ABSTRACT. The identification of Favella ehrenbergii, a marine planktonic ciliate, has largely been based on its lorica features. This approach is potentially problematic given the polymorphic lorica during this organism's life cycle. We isolated a population of F. ehrenbergii from the coastal waters of Incheon, Korea, and revealed its infraciliature using the protargol staining method. Phylogenetic analysis based on small subunit rRNA gene sequences was also performed. Results showed that this population possessed 16 collar membranelles (CM) and about 100 somatic kineties. These features are highly conserved, even in later dividers. As such, the number of CM and somatic kineties can be used as key characteristics for identification of Favella species.  相似文献

Calreticulin (CRT) plays pivotal roles in Ca2+ homeostasis, molecular chaperoning, infection, inflammation and innate immunity. In an attempt to study the involvement of CRT in innate immunity, the full-length cDNA of calreticulin (PxCRT) was cloned from the diamondback moth, Plutella xylostella. It consists of 1674 bp (excluding poly-A tail) with a longest open reading frame (ORF) of 1197 bp encoding 398 amino acids. In silico analysis of PxCRT ORF reveals that it has various repeat motifs and endoplasmic reticulum retention signal found in all the calreticulin proteins. As expected, high amino acid sequence identities were found from other CRTs identified from Bombyx mori (87%), Galleria mellonella (87%), Apis mellifera (74%), Anopheles gambiae (74%), Tribolium castaneum (73%), Culex quinquefasciatus (73%), Rhodnius prolixus (72%), Nasonia vitripennis (71%), Drosophila melanogaster (71%) and Haemaphysalis qinghaiensis (68%). During development, P. xylostella expressed PxCRT predominantly in the pupal stage. In addition, spatial expression pattern analysis indicates that PxCRT was highly expressed in the silk gland. PxCRT mRNA, furthermore, was strongly induced 3 to 6 h after laminarin treatment, suggesting that PxCRT appears to be involved in immune responses and also plays an important role in the silk gland.  相似文献

The perennial rhizomatous grass, Miscanthus×giganteus is an ideal biomass crop due to its rapid vegetative growth and high biomass yield potential. As a naturally occurring sterile hybrid, M. ×giganteus must be propagated vegetatively by mechanically divided rhizomes or from micropropagated plantlets. Plant regeneration through somatic embryogenesis is a viable approach to achieve large‐scale production of plantlets in tissue culture. Effect of the callus types, ages and culture methods on the regeneration competence was studied to improve regeneration efficiency and shorten the period of tissue culture in M. ×giganteus. Shoot‐forming calli having a yellow or white compact callus with light‐green shoot‐like structures showed the highest regeneration frequency. Percentage of shoot‐forming callus induction from immature inflorescence explants was 41% on callus induction medium containing 13.6 μM 2,4‐d and 0.44 μM benzyladenine (BA). The use of a regeneration medium containing 1.3 μM NAA and 22 μM BA was effective at shortening the incubation period required for plantlet regeneration, with 69% of total regenerated plantlets obtained within 1 month of incubation on regeneration medium. Embryogenic‐like callus morphotype could maintain regeneration competency for up to 1 year as suspension cultures. Field grown regenerated plants showed normal phenotypic development with DNA content and plant heights comparable to rhizome propagated plants. Winter survival rates of the regenerated plants planted in 2006 and 2007 at the University of Illinois South Farm, Urbana‐Champaign, Illinois, were 78% and 56%, respectively.  相似文献

This paper provides a review of the family Lucanidae in Korea, which consists of 17 species belonging to 9 genera. One new species, Dorcus tenuihirsutus sp. nov., is described and the following taxonomical changes are proposed: Aegus laevicollis Saunders, 1854 is newly identified as Aegus laevicollis subnitidus Waterhouse, 1873; Macrodorcas striatipennis Motschulsky, 1861 is removed from the Korean fauna because this record is thought to have been based on the misinterpretation of locality and misidentification; Neolucanus saundersii Parry, 1864 is removed from the Korean fauna because this record is thought to have been based on misidentification.  相似文献

Kim, J.K., Khim, B.‐K., Woo, K.S., & Yoon, S.H. 2009: Records of palaeo‐seawater condition from oxygen‐isotope profiles of early Pleistocene fossil molluscs from the Seoguipo Formation (Korea). Lethaia, Vol. 43, pp. 170–181. High‐resolution δ18O profiles of early Pleistocene fossil molluscs (Mizuhopecten tokyoensis hokurikuensis) from the shallow‐marine sedimentary Seoguipo Formation (Korea) were found to exhibit distinct annual cycles identified by their unique seasonality (δ18O amplitude). A direct comparison of fossil δ18O profiles with that of living shells (Amusium japonicum japonicumi) indicated that the palaeoseawater conditions differed from present‐day seawater. Specifically, the positive δ18O shift in the isotope profile of the fossil specimens relative to that of the living mollusc shell reflected that palaeotemperature was lower than that today. However, a comparison of the coldest palaeotemperatures (determined from the heaviest δ18O values of fossil shells), with the present‐day winter temperatures indicates that temperature variation alone cannot account for the entire positive δ18O offset. These findings indicate that variation in the seawater δ18Ow values plays a dominant role in the biogenic carbonate precipitation of fossils. Thus, the fossil shells obtained from stratigraphic units suggest different palaeoenvironmental conditions, including lower temperatures and 18O‐enriched glacial seawater, when compared with the present‐day conditions. The Seoguipo Formation records at least five cycles of relative sea‐level fluctuations, with changes in fossil δ18O amplitudes separated by sequence boundaries likely to reflect variations of unique palaeoseawater condition, although the oxygen‐isotope profile of fossil molluscs appears to provide a snap‐shot of the palaeoclimatic signature. □Early Pleistocene, mollusc fossils, oxygen isotope, palaeoenvironment, seawater temperature.  相似文献

Toxicity of 4-nonylphenol (4-NP) in the Chironomidae Chironomus flaviplumus was analyzed using a proteomics approach that involved identifying proteins by two-dimensional polyacrylamide gel electrophoresis (2-D PAGE). Proteome analysis of 4-NP-treated samples on silver stained gels found alterations in the expression levels of three proteins compared with control samples. Hsp70 proteins, so-called stress proteins, were studied in Chironomus flaviplumus exposed to different concentrations of 4-nonylphenol (0, 30, 100, 150, 300 and 600 mg/L) in the laboratory and in the field in captured animals from site 1 (1 km from a chemical factory) and site 3 (16 km from a chemical factory). Hsp70 proteins were found in all samples tested, including controls, but differed in their expression levels. At more polluted sites (site 1), the samples treated with higher concentrations of 4-NP more strongly expressed Hsp70. 2-D spots were induced or enhanced in gels following injection of 4-NP. Therefore, the induction of stress protein expression in Chironomus flaviplumus, in particular Hsp70, can be used as a biomarker for the evaluation of environmental conditions.  相似文献

The Plus ça change model predicts that deepwater trilobite species such as Triarthrus should exhibit gradual phyletic evolution. A detailed stratigraphic sequence of Triarthrus beckii specimens considered together with geographically separated samples from a single time interval provide a test of the Plus ça change model. We examined geographic patterns of variation in cranidial shape based on specimens from four approximately synchronous levels within the Upper Ordovician (lower Edenian) strata of New York, Québec, Kentucky and Pennsylvania. All geographic populations differ in mean size, except for New York and Pennsylvania. Because allometry is present in both meraspid and holaspid phases of the trilobite, size effects on shape were removed by ontogenetic standardization. Cranidial shapes were then compared among the four study sites and eight stratophenetic samples from New York, obtained by subdividing our Mohawkian Composite Standard Section into 20-m-thick intervals. Goodall's F-test of pairwise comparisons of cranidial shape between eight subintervals in New York and the other three populations are all significant. Geographic variation in cranidial size and shape in T . beckii appears to display a gradient or cline-like pattern that is related to paleogeography. Although cranidial shape is not entirely static within the New York stratophenetic series, it is in all cases different from that sampled elsewhere in the region. Thus, there is no evidence of wholesale immigration and emigration among the geographic areas. Instead, the geographic gradient of T . beckii shape variation may have remained fairly stable over ca. 3 million years during the mid-Chatfieldian to early Edenian interval. This result is indicative of stasis. Consequently, the evolutionary history of T . beckii contradicts the Plus ça change model.  相似文献
